Titre : Poly(ethylene glycol) in drug delivery, why does it work, and can we do better? All atom molecular dynamics simulation provides some answers. .

Résumé : Poly(ethylene glycol) (PEG) has an important role in drug delivery as a protective coating in pharmaceutical nanotechnology,  or as a covalently bound congugate of drug molecules. The mechanism through which PEG performs its function is not clearly understood. This is a highly relevant issue, as the search for a a superior alternative polymer to PEG is a very active field. We have used all atom molecular dynamics simulation to study this. We have performed simulations of the PEGylated liposome surface. We have studies the effect of liposome formulation, and interactions with salt ions in the bloodstream and targeting ligands on the liposome surface. We have also studied the interaction between PEG and three drugs for which it is used as a delivery aid: paclitaxel, piroxicam, and hematoporphyrin.
