RÉSUMÉ: Ipatasertib (GDC-0068) is a small-molecule Akt kinase inhibitor in late-stage clinical trials for oncology. We will describe our research and development efforts aimed at defining an efficient process to synthesize this complex active pharmaceutical ingredient. We will illustrate the interplay of process chemistry, organometallic catalysis, biocatalysis and API solid-state final form control considerations in the evolution towards an efficient route that bears the hallmarks of a commercial manufacturing process.
